- Measure 800 ml of distilled water in a beaker.
- Add 1.89 grams of Orchid Basal Salt, 30 grams of sugar, and PPMโข (1-2 ml/L) and stir the mixture thoroughly until all components are dissolved.
- Add distilled water to bring the total volume to 1000 ml.
- Adjust the pH of the solution to a range of 5.4 to 5.8 using a pH meter and appropriate pH adjusters.
- Add a gelling agent (6-8 g/L of agar or 3-4 g/L of Gellan Gum) to the solution while stirring continuously.
- Autoclave the prepared medium at 121ยฐC and 15 psi for 20 minutes to sterilize.
- Allow the medium to cool before pouring it into culture vessels.
