Murashige and Skoog Media (MS Media) is a nutrient medium designed to cultivate plant cells, tissues, and organs in vitro. Developed in 1962 by Toshio Murashige and Folke Skoog, it has become one of the most extensively utilised media in plant tissue culture.
MS Media contains a carefully balanced mix of macronutrients, micronutrients, vitamins, and plant growth regulators, providing the ideal environment for optimal plant growth and development.
Advantages and Applications of Murashige & Skoog Media
- User-friendly and straightforward to prepare.
- Supplies the essential nutrients plants need for in vitro growth and development.
- Offers a well-balanced formulation of nutrients, including vital minerals and vitamins, to support the cultivation of various plant tissues.
- Highly effective in promoting callus formation.
- Acts as a dynamic source of diverse compounds, including amino acids, organic acids, plant growth regulators (such as cytokinins and auxins), vitamins, sugars, minerals, nucleic acids, and other unidentified growth-promoting substances.
Steps for Using Murashige & Skoog Media
To prepare 1 litre of tissue culture medium using MS Media, follow these instructions:
- Fill a beaker with 800 mL of distilled water.
- Add 4.54 grams of PCT’s MS Media, 30 grams of sugar, and 1-2 ml of PPM™ (per litre) to the water.
- Adjust the total volume to 1000 ml.
- Set the pH of the solution to a range of 5.4-5.8.
- While continuously stirring, incorporate a gelling agent, such as 3-4 g/L gellan gum or 6-8 g/L agar.
- Autoclave the medium at 121℃ and 15 psi for 20 minutes.
Your tissue culture medium is now ready for use!
Important Notes:
- Always prepare the medium according to the specific formula provided on the label.
- For best results, using the entire container of MS Media at once is recommended.
- Heat-sensitive components should only be added after autoclaving.
